Purification of Tetragalloylglucose 4-O-Galloyltransferase and Preparation of Antibodies against This Key Enzyme in the Biosynthesis of Hydrolyzable Tannins

چکیده
The enzyme, ß-glucogallin: 1,2,3,6-tetra-O-galloyl-ß-D-glucose 4-O-galloyltransferase, which catalyzes the last common step in the biosynthesis of the two subclasses of hydrolyz able tannins, i.e. gallotannins and ellagitannins, was purified 868-fold from leaves of peduncu late oak ( Quercus robur, syn. Q. pedunculata) to apparent homogeneity. Polyclonal antibod ies against this pivotal enzyme were raised in rabbits and purified by protein-A chromatography, gel-filtration and affinity complexation. They were found to react specifi cally with acyltransferase from oak, displaying no cross-reactivity towards analogous enzymes from other plants synthesizing hydrolyzable tannins along the same biogenetic route, e.g. Rhus typhina or Tellima grandiflora.

Preparation and Purification of Polyclonal Antibodies against Mycobacterium Avium Paratuberculosis Antigens in Rabbit
Background and Objective: Johne’s disease is the chronic granulomatous enteritis of ruminants, and a major health hazard worldwide. In recent years, researchers have focused on mycobacterium avium subsp. paratuberculosis (MAP) antigens in diagnostic tests. Identification of antibodies against MAP antigens is, therefore, effective for the diagnosis or preparation of vaccine. The aim of this stud...

متن کاملPurification and Characterization of Alginate Lyase from Mucoid Pseudomonas aeruginosa Strain 214
Pseudomonas aeruginosa is an opportunistic pathogen that causes a variety of infections in compromised patients. The ability of Pseudomonas aeruginosa to produce chronic infection is based in part on its ability to biosynthesis of biofilm, and alginate is the major polysaccharide in the synthesized biofilm. So alginate degradation is very essential in the dispersion of Pseudomonas aeruginosa bi...
متن کاملProduction and purification of polyclonal antibody against F(ab')2 fragment of human immunoglobulin G
Antibodies are essential tools of biomedical and biochemical researches. Polyclonal antibodies are produced against different epitopes of antigens. Purified F(ab')2 can be used for animal’s immunization to produce polyclonal antibodies. Human immunoglobulin G (IgG) was purified by ion exchange chromatography method. In all stages verification method of the purified antibodies was sod...

Foot and mouth disease (FMD) is a contagious animal disease that causes irreparable damage to the economy of countries, including Iran in which this disease is a native one. Among the ways to combat FMD are vaccination and slaughter. Due to the specific situation of Iran, it is not possible to kill infected animals. Therefore, vaccination is the most important way to fight this disease. S...
